【摘要】:Stemcellsofhematopoieticoriginarepluripotentialandmaybeparticularlysensitivetotheeffectsofstimulationbynanoparticulatematerials.TheeffectofparticlesonmacrophageresponseshasanextensivehistoryandcanbeassessedbyPracticeF1903.Thetestmethoddescribedherewillassesstheeffectonstemcellswhichcanbeprogenitorcellstothemacrophageline.1.1Thistestmethodprovidesaprotocolforquantitativeanalysisoftheeffectofnanoparticulatematerialsinphysiologicsolutionongranulocyte-macrophagecolony-formingunits.1.2Thistestmethodemploysmurinebonemarrowhematopoieticstemcellswhichproliferateanddifferentiatetoformdiscretecellclustersorcolonieswhicharecounted.1.3Thistestmethodispartoftheinvitropreclinicalcharacterizationcascadefornanoparticulatematerialsforsystemicadministrationinmedicalapplications.1.4Thisstandarddoesnotpurporttoaddressallofthesafetyconcerns,ifany,associatedwithitsuse.Itistheresponsibilityoftheuserofthisstandardtoestablishappropriatesafetyandhealthpracticesanddeterminetheapplicabilityofregulatorylimitationspriortouse.
